The widely-used drug disulfiram metabolizes rapidly to form diethyldithiocarbamate, and it is impossible to study its metabolic rate inhibitory action on acetaldehyde metabolism without considering the properties of the metabolite, even though they have different spectra of pharmacological activity and different applications. Diethyldithiocarbamate is pharmacologically active in a greater number of ways than had been originally suggested. Its uses have ranged from an antidote to nickel carbonyl intoxication to the balancing of mossy fibre botons in the hypothalamus, and from its application as a rescue agent in cancer chemotherapy to its recently confirmed ability to inhibit the progression of HIV infections. There is sustained interest in its ability to chelate heavy metals, particularly cadmium, thallium and nickel, and whilst it continues to be used experimentally in cisplatin chemotherapy for protection against renal toxicity, there is now increasing interest in its ability to counteract bone marrow toxicity of a range of chemotherapeutic agents. There has been a recent increase in the use of disulfiram in the treatment of alcoholism, it having been credited with affecting a marked decrease in the incidence of cases requiring treatment for this condition. The growing level of research and clinical interest in disulfiram and diethyldithiocarbamate has resulted in a need for an integrated synthesis of current knowledge on the subject. This book provides a critical review of the current status of research in the properties, pharmacology and toxicology of disulfiram and diethyldithiocarbamate and their use for the treatment of a variety of human disease conditions, including AIDS, cancer, alcoholism and heavy metal intoxication. The text is supported by a bibliography and the book should serve as an important reference source for pharmacologists, biomedical scientists, immunologists, biochemists and clinicians.

Añadir al carritoTaschenbuch. Condición: Neu. Druck auf Anfrage Neuware - Printed after ordering - This book is aimed at those in the biomedical community that are interested in the therapeutic applications, pharmacology, biochemistry, toxicology and pharmacokinetics of the title compounds. Recent findings regarding the ability of diethyldithiocarbamate (ditiocarb, Imuthiol®) to delay the progression of HN infections and AIDS, the discovery of its potential as a rescue agent in cancer chemotherapy, and the identification of disulfiram (Antabuse®) regimens that allow alcoholics to achieve abstinence of many months' duration have made writing this book an exciting experience. At the same time the fact that the two drugs differ substantially in their pharmacological effects in spite of their easy interconvertibility has rendered the writing intellec tually challenging. Diethyldithiocarbamate, an agent seemingly less toxic than aspirin, rivals it in the multiplicity and diversity of its pharmacological proper ties. Notable among these are the manyfold potent immunostimulant effects, and though most of these involve effects on T -cells, the mechanism of diethyldithiocarbamate's action remains far from clear. The drug is also a potent chelator of heavy metals and this has led to a number of clinical applications. As might be expected, it inhibits several important enzymes. Further, it is one of the most effective radioprotective agents; it also protects organisms against a variety of toxic agents.

Most ore deposits that were mined in the past or are currently being extracted were found at or near Earth's surface, often serendipitously. To meet the future demand for mineral resources, exploration success hinges on identifying targets at depth. Achieving this requires accurate and informed models of the Earth's crust that are consistent with all available geological, geochemical and geophysical information, paired with an understanding of how ore-forming systems relate to Earth's evolving structure. Contributions to this volume address the future resources challenge by (i) applying advanced microscale geochemical detection and characterization methods, (ii) introducing more rigorous 3D Earth models, (iii) exploring critical behaviour and coupled processes, (iv) evaluating the role of geodynamic and tectonic setting and (v) applying 3D structural models to characterize specific ore-forming systems.
