Today all known steroid hoITIDne receptors have been cloned and sequenced and novel members of this supergene family are constantly being discovered. Sane of these new receptors remain to be characterized regarding the nature of their ligand. Access to probes for steroid receptors has enabled studies on mechanisms of regulation of receptor gene expression. Deletion and II'Altational analysis of steroid receptor cDNAs followed by expression in cells together with suitable reporter genes has yielded a detailed knowledge about the functional significance of the various domains the receptors are composed of. In certain cases, steroid resistance in patients have been shown to be due to point mutations in the corresponding steroid receptor genes resulting in non-functional receptors. The availability of receptor cDNAs also makes it possible to express receptors at high levels in procaryotic and eucaryotic cells. It is e. g. possible to express the DNA-binding domain of the glucocorticoid receptor in E. coli as a fusion protein with protein A which interacts specifically with DNA. Such studies are necessary for production of sufficient quantities of receptors to allow crystallization and X-ray crystallography for detailed structural information.

Initial interest focussed on the actions of the ETs in the cardiovascular system, and this remains the pri mary source of published work involving these peptides. Importantly, evi dence is mounting for a significant mediator role for ET-l in cardiovascu lar diseases including systemic hypertension and congestive heart failure. However, it was also recognized early on that the ETs exert an array of powerful actions in the respiratory tract, many of which prompted specula tion concerning mediator roles for ET-l in several lung diseases including asthma and pulmonary hypertension. In recent years the evidence for the involvement of ET-l in lung diseases has become more compelling. There is now real hope and anticipation that novel ET receptor antagonists may eventually be used therapeutically against asthma and other major lung pathologies. It is this atmosphere of excitement created by new discoveries and the increasing scientific ratio nale for a significant impact ofthe ETs in pulmonary diseases that prompt ed the creation of this book, the first on this topic. In the past decade both the number of workers and their publications has grown at some… thing approaching an exponential rate and, more importantly, the field has gradually expanded to encompass a large number of diverse areas (mediators, receptors, cell effector mechanisms, regulatory factors, etc.). It is now apparent that leukocytes are particularly useful for studies of the locomotory behavior of all cell types and of mechanisms controlling cell movement and orientation. Chemotactic factors, once discovered as substan ces able to induce directional migration in leukocytes, are now recognized as potent stimulators of a variety of cell functions. Based on our knowledge in the field of basic research in leuko cyte chemotaxis, clinical observations in combination with ex perimental studies in vivo have provided new insights into the role of leukocyte-mediator interactions. The recognition that a specific interaction between chemotactic factor and leukocyte can lead to a multitude of cellular responses and products has opened up a broad area of study. General features of the specificity of dehydrogen ase reactions are described. The pyridine nucleotide - disulphide oxidoreductases exemplify a type of multicomponent enzyme complex. Lipoamide dehydrogenase, glutathione reductase, and thioredoxin reduct ase are described and compared. The fatty acid synthesizing systems of some organ isms are nonaggregated, whereas in other organisms they are multienzyme complexes containing, in animal tissues for example, two closely related multifunctional polypeptide chains. The dehydrogenase activities of the fatty acid syn thesizing systems afford many points of interest, which are described and discussed. Hydroxymethylglutaryl-CoA reductase occupies a key position in the synthesis of isoprenoid compounds, including cholesterol. It probably occurs in all forms of life; in animal cells it is a microsomal enzyme. Its properties are reviewed. 288 pp. Englisch.

Als Tagungssekratär wirkte Herr Ger (Ka.towice). Die :{ Teilnehmer kamen aus Europa, Amerika und Australien. Erfreulicherweise hat sich eine grtlßere Zahl von Teilnehmern aus Polen eingefunden; leider konnten keine Wissenschaftler aus Jugoslawien, Rumänien, der TBchechoslawakai und Ungarn kommen. Die Tagtmg wurde von Herrn Aczel eröffnet und von Herrn Aumann geschlossen. Zu der durch den Titel gegebenen allgemeinen Thematik haben die Teil nehmer in sehr verschiedene Weise beigetragen. Trotzdem bildeten sich einige Schwerpunkte heraus: Funktionalungleichungen, inbesondere iterativen ~s, konvexe und verallgemejnert konvexe Funktionen, Differentialungleichungen, Ungleichungen der Funktionentheorie, Funktionalanalysis und Geometrie. Ferner wurden Anwendungen von Ungleichungen auf Differentialgleichungen, Physik, Warscheinlichkeits-, Informations- und Wirtschaftstheorie gebracht. Die als besondere Tagungspunkte augesetzten 'Bemerkungen und Probleme' haben sich als äußerst fruchtbar erwiesen, ja sogar in einem Fall zur vollständigen LÖsung eines aufgeworfenen Problems geführt. Die Tagung hat nicht nur gezeigt, daß das Netz von Ungleichungen, das die. gesamte Analysis durchzieht, verdient in eigenständiger Weise und im Sinne allgemeiner Methoden behandelt zu werden, sondern auch bestätigt, wie viel schneller und wirksamer der Ideenaustausch ist, der durch die Vorträge und durch den persönlichen Kontakt der Wissenschaftler untereinander zustande kommt, als die unpersönliche, nur literarische Information. 352 pp. Englisch.

The primary aim of this book is to give an approach which would be a…s intelligible and lucid as possible. Our definition, produced in Chapter I, requires for its background only a little of the theory of absolutely convergent series so that it is understandable for students of the first undergraduate course. Nevertheless, it yields the Lebesgue integral in its full generality and, moreover, extends automatically to the Bochner integral (by replacing real coefficients of series by elements of a Banach space). It seems that our approach is simple enough as to eliminate the less useful Riemann integration theory from regular mathematics courses. Intuitively, the difference between various approaches to integration may be brought out by the following story on shoemakers. A piece of leather, like in Figure 1, is given. The task consists in measuring its area. There are three shoemakers and each of them solves the task in his own way. The sum of areas of all rectangles is taken for an approximate area of the leather (Figure 2). This volume together with the earlier volume 'Contributions to Geometry' edited by Tolke and Wills and published by Birkhauser in 1979 should give a fairly good account of many of the more important facets of convexity and its applications. Besides being an up to date reference work this volume can be used as an advanced treatise on convexity and related fields. We sincerely hope that it will inspire future research. Fenchel, in his paper, gives an historical account of convexity showing many important but not so well known facets. The articles of Papini and Phelps relate convexity to problems of functional analysis on nearest points, nonexpansive maps and the extremal structure of convex sets. A bridge to mathematical physics in the sense of Polya and Szego is provided by the survey of Bandle on isoperimetric inequalities, and Bachem's paper illustrates the importance of convexity for optimization. The contribution of Coxeter deals with a classical topic in geometry, the lines on the cubic surface whereas Leichtweiss shows the close connections between convexity and differential geometry. The exhaustive survey of Chalk on point lattices is related to algebraic number theory. A topic important for applications in biology, geology etc. 424 pp. Englisch.

During the past decade a large number of new compounds were marketed, some of which had to be withdrawn after a short… while because of adverse drug reactions. These experiences led to an under standing that a complete picture of the safety of these drugs can only be obtained after their introduction into the marketplace. Differentmethods ofpost-marketing surveillance (PMS) serve as important tools for monitoring the frequency of adverse reactions and for generating and corroborating hypotheses. Experience with PMS has thusfar been quite limited in Germany. In 1987 a non-profit organization, 'Verein zur Langzeituntersuchung von Arzneimittel wirkungen auf dem Gebiet der Rheumatologie e. V. ' (VLAR) was founded by interested physicians and pharmacologists to raise funds and perform investiga tions on the safety of NSAIDs. The first project by the VLAR, SPALA ('Safety Profile of Antirheumatics in Long-Term Administration'), was sponsored by F. Hoffmann-La Roche AG (Basel, Switzerland). In July 1990, when the project was successfully terminated, almost 30,000 patients had been completely docu mented and their medical records entered into a computer for subsequent review by a select panel of experts with experience in monitoring adverse reactions to NSAIDs. The comments, criticisms, and ideas of these experts were brought togetherat a symposium organized by the VLAR at the Klinikum Steglitz (Berlin, Germany) on 12 October 1990. 116 pp. Englisch.
