Son of a wealthy Florentine merchant, he turned away from a commercial career to take up scholarship under the guidance of the great civic humanist, Leonardo Bruni. Like Bruni he mastered both classical Latin and Greek, but, unusually, added to his linguistic armory a command of Biblical Hebrew as well. He used his knowledge of Hebrew to make a fresh translation of the Psalms into humanist Latin, a work that implicitly challenged the canonical Vulgate of St. Jerome. His Apologeticus (1455-59) in five books was a defense of the study of Hebrew and of the need for a new translation. As such, it constituted the most extensive treatise on the art of translation of the Renaissance. This ITRL edition contains the first complete translation of the work into English.

El Apologeticus constituye una encendida defensa de la traducción latina del Salterio hebreo, que Giannozzo Manetti llevó a cabo en el marco de un ambicioso proyecto encargado por el pontífice Nicolás V: verter al latín todos los textos sagrados desde sus originales hebreo y griego. La versión de Manetti ni siquiera llegó a ver la luz, lo que no impidió que aparecieran numerosos detractores que argumentaban la futilidad de tal empresa ante la existencia de las dos versiones anteriores de San Jerónimo, una sobre el texto de la Septuaginta, otra sobre el original hebreo. La gran aportación de Manetti radica en que aplica el concepto de tipología textual a la teoría de la traducción. Frente a los planteamientos de Leonardo Bruni y otros humanistas del momento, Manetti defiende la necesidad de que el traductor se adapte a la particular idiosincrasia de los textos religiosos y filosóficos, a partir de una vía intermedia entre las versiones ad verbum y ad sensum.
