The objective of this conference was to provide a unique opportunity for scholars working in these two related and rapidly growing fields to participate in the exchange, integration, and synthesis of new concepts and ideas, to engage in constructive criticism and to initiate new collaborative research projects. The conference focused on the molecular and cellular aspects of aging as related to oxidative stress. It was one of the largest and most comprehensive international conferences held in molecular gerontology. However, in many countries their popularity is waning as prescribers become more aware of problems with unwanted effects and with rebound and withdrawal syndromes. The earlier benzodiazepines were long-acting, and it was thought that their shorter-acting successors would have important advantages. Other developments include partial agonists and partial inverse agonists, compounds acting on the benzodiazepine-GABA macro molecular site although not as a typical benzodiazepine, and anxiolytic agents acting on 5-HT pathways directly. I shall review the problems of unwanted effects and dependence, mainly from an epidemiological view point, and then relate the pharmacological properties of the newer com pounds to possible differences in their clinical profile of adverse effects. Adverse behavioural effects Benzodiazepines are used, inter alia, as anxiolytics and hyp notics. The latter use exploits the sedative actions which occur at higher than anxiolytic doses. The sedation should disappear by the time the patient wakes up. It is probably more efficient to present such material after a reasonable competence in (at least) linear algebra and calculus has already been attained - but at this stage those not specializ ing in numerical mathematics are often interested in getting more deeply into their chosen field than in developing skills for later use. An alternative approach is to incorporate the numerical aspects of linear algebra and cal culus as these subjects are being developed. Their existence was implied by Fatt and Katz in 1952, when they described the action of acetylcholine at the muscle endplate as producing 'a large nonselective increase of ion permeability, i.e. a short circuit'. In the last two decades, some extensions of the finite difference method to irregular networks have been described and applied to solving boundary value problems in science and engineering. For instance, 'box integration methods' have been widely used in electro nics. There are several papers on this topic, but a comprehensive study of these methods does not seem to have been attempted. The purpose of this book is to provide a systematic treatment of a generalized finite difference method on irregular networks for solving numerically elliptic boundary value problems. Thus, several disadvan tages of the classical finite difference method can be removed, irregular networks of triangles known from the finite element method can be applied, and advantageous properties of the finite difference approxima tions will be obtained. The second international conference on 'New Drugs in Allergy and Asthma' was held in Davos on 8-10 July 1992. There were six sessions with specialist chairmen: PDE isoenzymes (c. D. Nicholson), atopy and the environment (8. Villiger), adhesion molecules (R. H. Gundel), cytokines (M. Baggiolini), airway hyperreactivity (S. Makino) and neurotransmitters (J. G. Widdicombe). The success of the meeting was ensured by a high standard of presentation, with extensive reference to recent experimental data. The chairmen were instrumental in fostering stimulating discussion periods, that considerably enlivened the meeting. It is the third meeting in the series started three years ago in which we tried to put together mathematical physicists from the member and non-member countries of JINR with their colleagues from soviet universities and institutes using this international centre as a convenient basis. As in the previous cases, new faces, subj ects and ideas appeared but the spirit remained the same, relaxed and inspirative. During the last few years definite advancement has been made in elucidating the molecular and genetic aspects of the enzyme involved in oxidative phosphorylation, the so-called FoFl H~-ATP synthase. Non-invasive NMR technologies have been developed to monitor in vivo the energy level of tissues based on determination of the concentrations of adenine nucleotides, phosphate and phosphate esters. Thus it became clear that the capacity of oxidative phosphorylation adapts itself to the ATP demand which changes continuously with the physiological state in various tissues. This is achieved by regulation of the enzyme activity as well as by regulation of its biogenesis. The reversible phosphorylation of proteins is re cognised as a major regulatory mechanism in eukaryotic as well as in prokaryotic cells for cellular signal transduction and control of gene expression, cell growth, differentation and oncogenesis. The same applies to the role played by cAMP. A further topic of growing interest concerns the discovery of the ATP binding cassette (ABC) superfamily of transport proteins which includes systems of primary importance in medicine such as the multi-drug resistance P glycoprotein, the cystic fibrosis transmembrane conductance regulator (CFTR) and the 70 kd peroxisomal membrane protein. The classical anti-histamines, now known as H -antagonists have 1 been the result of intensive research in the 1950s and 1960s. Polya, who had been the last surviving author of the book InequaZities (Cambridge University Press, 1934), who died at the age of 97 years and whose many and manifold contributions to mathematics will be recorded elsewhere, in due course. Neuware -Uncontrolled proliferation of vascular smooth muscle cells (SMC) in response to vessel injury is a problem with a considerable therapeutic impact. Specifically, restenosis after percutaneous transluminal coronary angioplasty (PTCA) is a clinical problem without any effective drug therapy so far. Thus, there is need for an improved drug therapy but also for an improved understanding of the pathophysiology of growth control in SMC. Cyclooxygenase products, such as prostaglandins and thromboxane, are intimately involved in growth responses. Vasodilatory prostaglandins, such as PGI , PGE or their analogues, have 2 1 been shown to inhibit SMC proliferation. There is also evidence for a markedly increased endogenous prostaglandin production during neointirna formation under the influence of growth factors which includes induction of COX-2. These data suggest that prostaglandins might be considered both targets and tools of growth control. The application of DNA finger printing to forensic and legal medicine has guaranteed a high public profile for this technology, and indeed, scarcely a week goes by with out the press reporting yet another crime successfully solved by molec ular genetics. Less spectacularly, but equally importantly, DNA typing methods are steadily diffusing into an ever wider set of applications and research fields, ranging from medicine through to conservation biology. To date, two DNA fingerprinting workshops have been held in the UK, one in 1988 organised by Terry Burke at the University of Leicester, and the second in 1989 at the University of Nottingham, co-ordinated by David Parkin. In parallel with these workshops, which have provided an important focus for researchers, Bill Amos and Josephine Pemberton in Cambridge have established an informal newsletter 'Fingerprint News' which is playing a major role as a forum for DNA fingerprinters. By 1989, it was clear that the field had broadened sufficiently to warrant a full international meeting. As a result, Gaudenz Dolf took on the task of organising the first, of what I hope will be many, International Symposium of DNA Fingerprinting held at Bern during Ist-3rd October 1990.
